Alignment as evolution's reflection.

Alignment of homologous proteins sequences

A 6-sequence alignment was built with proteins chosen from HSP70 family from Eukaryota, Bacteria and Archea. The coloured alignment by ClustalX scheme with Identity Threshold = 100% condition is presented on picture. The colour scheme is such that only coloumns with 100% identical aminoacid residues in each sequence are coloured.

A markup Labels was added. Letter C marks positions, which are more than 80% conservative, F is for functionally conservative ones and G means gaps. There are valine, leucine and isoleucine (hydrophobic amino acids) on positions 69,77 and 78.

Main properties of this alignment are presented in the following table:

ID 100% conservative % Gaps % Functionally conservative % 70% conservative %

Fulfilled corrections:

  • On positions 20, 21, 22 serine and valine were replaced to the left, because deletion and insertion are more feasible than mutation.
  • On positions 120 and 121 the same thing was done with leucine.
  • On positions 228 229 leucine and glutamic acid were also replaced to the left.

Mutations order:

  • On position 195 occured phenylalanine insertion (between original sequence and ancestor 1)
  • On position 6 occured alanine insertion (between ancestor 4 and 5)
  • On position 45 occured mutation from arginine to cysteine (between ancestor 5 and 6)
  • On position 159 occured delecion of aspartic acid (between ancestor 1 and 2)
  • On position 20 occured serine insertion (between ancestor 3 and 4)
  • On position 31 occured lesine insertion (between ancestor 4 and 5)
  • On position 88 occured alanine insertion (between ancestor 6 and 7)
  • On position 207 occured threonine deletion (between ancestor 4 and 5)
  • On position 209 occured mutation from glycine to asparagine (between ancestor 6 and 7)
  • On position 130 occured isoleucine insertion (between ancestor 2 and 3)
